La Mrs. Robinson del titolo è un personaggio del film Il laureato di Mike Nichols, a cui il brano fa da colonna sonora. Nella pellicola Benjamin Braddock - il protagonista interpretato da Dustin Hoffman - è un giovane di buona famiglia, che intreccia una relazione con la signora Robinson, moglie del socio di suo padre.
La canzone vinse un Grammy Award nel 1969 come Record of the Year e fu il secondo singolo di successo di dopo "Sound of Silence".
